Résumé
Precise editing of human hematopoietic stem an progenitor (HSPC) is of interest both for the treatment of genetic disorders and for disease modeling. Current strategies using CRISRP/Cas9 with adeno-associated virus (AAV) vectors achieve in the range of 10-20% efficiencies on average, thus requiring additional selection for effective use.
In this lecture, I’ll go over a recent optimization performed in my lab which has allowed us to routinely achieve >90% precise editing efficiencies, including using short oligonucleotide donors. I will also discuss some of our recent results applying this system to the study of SRSF2 P95H and SF3B1 K700E mutations.
